There is no absolute answer to this question, as it depedns on many, many varying factors, but if you had to go with a gut reaction and say one or the other, which would it be?

Which city if more expensive to live in : Perth or Brisbane?

Just a simple one word answer.....

Its expensive here in Perth thats for sure.

i know the OP wanted one word answers but it isnt that easy
i based my answer on the median house price as the perth median is $50k dearer than brissie ( using realestate median figures from the internet )

and as your house is without dout the biggest ivestment / cost in the cost of living ive answered perth

Brisbane - but where ever you go be prepared for big prices and big interest rates and a constant struggle (ignore if your minted already). We a havent got kids and are both working and are paid good money, yet at the end of the month the cupboard is bare!! If we wanted to go home for an emergency tomorrow we couldnt, simple as that.... As my Wife says 'the sunshine doesnt pay the bills'.

House prices are creeping up on the coast here whereas on the way down in Perth.

Food esp. fruit and veggies is more expensive in QLD-ironically.

Flights are cheaper

So suppose, it all weighs up !
