The good news is that going from Mississauga to Kitchener means you're going against the massive flow of traffic that heads into the Toronto area every morning, so it's a relatively 'known' time to do the commute. If you were heading into the Toronto area from west of the city, you'd really have a lot of variation on how long your commute would be - but it's always a busy commute into the city.
Depending on what your significant other does for a living, I'd explore a bit before committing to a particular location - imho, there's a definite difference in 'feel' between Kitchener and Mississauga - which area suits you 'better' will all depend on what you're hoping to get out of your experience by coming to Canada.

As mentioned above, there's really not a good option for commuter train service from Mississauga to Kitchener at the moment, so it's a drive west on the 401.

There's no downtown in Mississauga. It's truly a place where there's no there, there. Port Credit and Streetsville have a few shops and restaurants but they're like Oakville, relatively nice looking dormitory locations.

Hi there, we are in the same boat, just waiting for the go ahead from my husbands company to transfer from UK to Kitchener and so now frantically looking online at areas that would be suitable for us. We have childen ages 4 and 9 so schooling will be an important factor for us too. Feel free to get in touch if you would like and we can swap ideas etc

The Downtown in Mississauga is basically Square One shopping Mall with a few things dotted around it.
If I could twin Mississauga it would be with Milton Keynes.
I think if one wanted to position themselves to work in Mississauga and Kitchener then Milton must be the destination that comes into play. That's even less of an eye pleaser.
