Do the Rome and Pisa train stations have lockers that can be rented?
About how much?

Also, will a large backpack fit in them? If not, what is another
option for storing a backpack for a day?

All lockers in Italy were closed post 9/11 due to 'security concerns'.
But Rome's Termini train station and Pisa's Centrale train station both
have manned luggage storage (deposito bagagli).
Termini's is downstairs (elevator) alongside Track 24 and Pisa's is
outside alongside Track 1 (on arriving turn right on Track 1).
Size doesn't matter:), if they can pick it up, they will store it.
I don't recall the cost but guessing, I'd say ~ €4-5 for 12hrs, the
FCO airport charges €3.5/24hrs but I know the train stations prices

I was at Pisa the other day - there was certainly a sign to a
left-luggage facility.

There was a long queue in front of the left luggage counter when I last used
it. Every piece of luggage had to be screened, and we needed to wait for
about half an hour...

Luggage lockers are more convenient, but are considered a security risk
nowadays... Koeln Hbf has a neat solution to the dilemma though.

Here's how it works: there are 9 consoles (looking a bit like oversized
ticket vending machines). You walk to one available, pay for a days
storage, and a little gate opens near the bottom where you'll find a box
you can put your stuff in. After it closes the box is whisked away to
underground storage, and you receive a magnetic card in exchange.
Put your card in any of the consoles and after a few seconds you get your
luggage back...

Sounds like an interesting solution. For security, but also spacesaving.
Lockers at big stations usually occupies large areas typically with long
dull, grey corridors.
