from Oct' 09 i have accepted a position in some other organization as a project manager but in the same industry (i.e software development).

I am almost certain that no it will not be a problem for the visa app, 12 in last 24 months is the requirement. I wasn't working at all when I applied.

Do you already have skills assessment?

No, you do not. So long as you have 12 of the last 24 months you should be fine. As Bermudashorts said, she was not even working when she applied; only had the requisite number of months experience.
