In brief, it has to be an approved offer with HRDC, meaning it meets
certain requirements, such as it can't be filled by a Canadian.

Re: Job Offer can get me Work Permit?
You need to have job offer validated by HRDC - it is employer's
responsibility to apply for such validation and to prove to HRDC that
despite extensive national search no Canadian candidate was found. Depending
from the nature of the job, your qualifications and nationality you may
qualify for work permit process that doesn't require individual validation.
You didn't provide any information about offer, employer and your
qualifications as well as nationality, thus it is impossible to tell you
more. Visit CIC website to learn more about work permit process and
available options here:
http://www.cic.gc.ca/english/work/index.html
Most of the time (when offer and/or yourself don't qualify for exemption
from individual validation) letter offering you a job but not validated by
HRDC is worthless, sorry.

Re: Job Offer can get me Work Permit?
Also note, there are certain HRDC exempt jobs, for which getting a
workpermit is relatively quick and easy ( about 40 days)

I think also it will depend on what type of HRDC validation you receive there are different types: The one I have is what is known as a Arranged Employment Opinion (AEO) and in the words of HRDC I quote:
"Please note this confirmation does not authorise the foreign national to enter, remain or take employment in Canada, nor can it be used to apply for a work permit"
Anyway good luck with you application

Re: Job Offer can get me Work Permit?
Pad:
Take a look at the web page provided by Jim as a job offer in Canada is only
valuable if it is certified, or
you fall within one of the specialized 7 software development occupations or
you fall within one of the reciprocal programs like NAFTA North
American Fee Trade and you have a job offer and a profession identified in
the specific program. If not, you would have to have
your employer obtain a HRDC (Human Resource Development Canada, now called
HRSD Human Resource Skill Development) certification or opinion about the
job(a process to determine the availability of Canadian residents qualified
and ready to do the job). A certification would allow you to come to Canada
to work while the opinion would give you an additional 15 points in the
selection grid in an application as a skilled worker.
