Question In my circumstances, which is best way to apply for a Work Permit ?

Hi, first time here, but finding it very interesting so far.

I am currently living in Canada on a Temporary Resident Permit as a Visitor from UK.

I have until Dec of this year left. Since I have had approval from HRDC with a Job Offer, I am unsure of which would be the best way of moving forward without 'messing up'.

I have a number of questions, which I hope someone could help with.

Can I apply for the Work Permit whilst here in Canada?
Should I apply to London or can I apply to Buffalo?
Subject to recieving the Permit, can I re-enter through Buffalo?
Thanking you all in anticipation
