Could anyone help me as to whether if myself and my wife are both on the skills list we would be better to get two separate visas or one and a spouse visa?

To apply for a Spouse visa one of you must be a citizen or permanent resident.

One of you needs to apply for a skills visa, then the other is listed onthat as a dependent.
