Is this journey covered by train? If so, how long is the journey and
do you need to change trains? If not, is there a regular bus service?

The local bus company based in Huelva is called DAMAS. They run busses all
over that province.
On the website http://www.damas-sa.es they advertise bus from Matalscanas
to Seville at 10.30 & 15.00 (Mon to Sat) or 16.00 & 19.00 Sunday &
'Festivos' (which are 'express' services, and there is a daily one at
18.00.
You can get the return times as well.
I hope that this website is up to date !
But surely there are busses into Huelva and from Huelva bus station there
are regular ones into Seville.
