First of all
you will have to buy tollroads stamps
in hungary I remember some highways were free. some were with
tollroadsticker some were with toll
to pay at the exit
the best route would be
Prague
Brno
Bratislava
Komarno Budapest
but that depends onw where you want to go.
I suggest the thermal bath of Heviz and the town of Eger.

Then he'll also have to buy stickers for CZ and SK.

This doesn't stop it being the best direct route though.
But it does miss out Cesky Krumlov?

Is is however not clear to me whether the request is
for places to see or just general things to be aware of
about driving in those countries.

this route is very far from Cesky Krumlov
in fact.. the best route to Cescky Krumlov would be
Prague
Ceske Budejovice
Cesky Krumlov.
then it would be better to take the road :
ceske budejovice freistadt linz vienna budapest ( all toll road in
hungary )
the road to C.K. is a normal road and it would take about 3 hours to reach
it
with
www.viamichelin.com it would be possible to make itineraries
