I am canadian citizen and my family ( wife and kid ) have got PR status of Canadian , however we have moved back to Pakistan and would like to take the US visit visa of my family ( wife and 4 year kid ). Now my question is should we apply for the US visa from Pakistan or apply from Canada.

This is Canadian immigration forum and you are asking about US visa, thus nobody cares to answer.
Americans don't care about your Canadian PR, especially when you moved back to Pakistan and have no ties to Canada.
Your nationality is what counts for Americans and substantial ties to your home country will be required if you want a chance for a visa. And of course you apply for visa in the country of your nationality and residence - as you stated you moved back to Pakistan, then it is obvious that Canada is not where you apply for US visa.
